If you are talking about for fair use, such as uploading a cover of your own for non-commercial use, than yes. But if you plan to record the material and sell it, you have to have permission from the copyright holder before you can profit off of it. The only exception for that is Parody. You can parody a song without having to get permission (see "Weird Al"), but it is still good to have their permission for his (once more, see "Weird Al").
